Follistatin 344 (FST-344)
Follistatin (FST) is a secreted glycoprotein that was first identified as a substance that inhibits follicle-stimulating hormone in the follicular fluid of the ovary (1, 2). The cDNA of human follistatin encodes a protein of 344 amino acids (aa) with a signal sequence of 29 aa, a non-terminal atypical TGF-binding domain, three follistatin domains that contain EGF-like and kazallic motifs, and a highly acidic tail of the terminal. Follistatin is a secreted protein that binds to ligands of the TGF-Beta family and regulates their activity, suppressing their access to signaling receptors. Initially, it was discovered as an activin antagonist whose activity suppresses the expression and secretion of the pituitary hormone FSH (follicle-stimulating hormone). In addition to the fact that it is a natural antagonist, follistatin can inhibit the activity of other TGF beta ligands, including BMP-2, -4, -6, -7, Myostatin, GDF-11 and TGF-Beta1. Follistatin is expressed in the pituitary gland, ovaries, decidual cells of the endometrium and in some other tissues. Recombinant human follistatin is a 37.8 kDa protein containing 344 amino acids.
