Previous work in our laboratory prompted us to study the effect of short-term, acute administration of TRGyl on organs, tissues, and aging-related metabolic and hormonal markers in order to gain more knowledge about the effects, dosage, and timing of administration in accordance with its circadian cycle. In addition, we wanted to test its antiaging effect on two of its most fundamental functions, namely: gonad-reproductive and renal-urinary. The results show that both short-term, acute or chronic, long-term oral administration of TRH to old, aging mice leads to positive changes and rapid correction to more juvenile levels of the most typical hormonal and metabolic changes associated with aging.

It is noteworthy that 4-month oral TRH treatment maintains testicular function in aging mice. As can be seen from the significant increase in the weight of the testes, TRH taken from drinking water provides maintenance and/or restoration of the structure and function of the testes, which is manifested in the active proliferation and formation of mature spermatogonia and intensive spermatogenesis in the follicles. 4-month oral TRH treatment protects the kidneys from amyloid and hyaline infiltration of both tubules and glomeruli, which is typical for aging mice. In fact, massive deposits of amyloid and hyaline material clearly penetrate the dried glomeruli of untreated mice with loss of filtering ability, while they are almost absent in mice treated with TRH. Massive hyaline degeneration can also be observed in tubular vessels of untreated control mice.

These experiments with parenteral and oral administration of TRH show the most remarkable effects of the neuropeptide TRH, which slow down aging and, apparently, even reverse aging. Again, like melatonin, we are faced with an antiaging agent with a wide spectrum of action, which must necessarily be associated with the most fundamental role in the regulation of metabolic and hormonal functions. Properties of aging-reversing thyrotropin-releasing hormone (TRH) What is it? TRH? TRH, also known as "thyrotropin-releasing hormone", bears the unfortunate and very misleading designation of thyroid-releasing hormone. It is a ubiquitous tiny molecule consisting of three amino acids, and it can be found everywhere in nature, from unicellular algae to complex mammalian species.
