Triptorelin (GnRH)
Triptorelin, a decapeptide (pGlu-His-Trp-Ser-Tyr-D-Trp-Leu-Arg-Pro-Gly-NH2), is a gonadotropin-releasing hormone agonist (GnRH agonist), used as acetate or pamoate salts. luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Like other GnRH agonists, triptorelin can be used in the treatment of hormone-sensitive types of cancer, such as prostate cancer or breast cancer, premature puberty, estrogen-dependent conditions (such as endometriosis or uterine fibroids), and also in assisted reproduction. Triptorelin is sold under brand names Decapeptyl (Ipsen) and Diphereline and Gonapeptyl (Ferring Pharmaceuticals). In the United States it is sold by Watson as Trelstar. In Iran, triptorelin is sold under the brand name Variopeptyl (Varian Darou Pajooh). In India, Dr. Reddy's Laboratories recently launched triptorelin under the brand name "Pamorelin LA". During the treatment of prostate cancer, it causes a surge in testosterone (the initial level of testosterone), known as the flare effect. In men, the drop in serum testosterone to the level normally seen after surgical castration occurs approximately two to four weeks after the start of therapy.
Triptorelin increases the level of testosterone, causing constant stimulation of the pituitary gland, reduces the secretion of gonadotropins, luteinizing hormone (LH) and follicle-stimulating hormone (FSH) by the pituitary gland. Like other GnRH agonists, triptorelin can be used in the treatment of hormone-sensitive types of cancer, such as prostate cancer or breast cancer, premature puberty, estrogen-dependent conditions (such as endometriosis or uterine myoma), as well as during assisted reproduction.
